Article Title: Effects of crude oil on juvenile threespine stickleback somatic and immune development
doi: 10.1101/2021.11.17.469020
Figure Lengend Snippet: This workflow was used to determine the effects of crude oil on juvenile threespine stickleback. Fish were reared in culture flasks from 1-14 dpf and in tanks from 14-28 dpf. Alaska North Slope crude oil or untreated water was added to randomly assigned flasks at seven dpf. Samples of 12 fish/flask were taken at 14 and 28 dpf. Fish were imaged, guts were dissected out (DNA yield too low for any analysis), and the whole bodies of three fish (from the same flask) were pooled for RNA isolation and qRT-PCR.
Article Snippet: Each sample was analyzed in triplicate qRT-PCR reactions using the Luna Universal One-Step RT-qPCR kit (New England Bio Labs, Ipswich, MA, USA) according to the manufacturer’s protocol.
Techniques: Isolation, Quantitative RT-PCR
